Presents a compilation of HPLC methods for the separation and quantitation of porphyrins and porphyrin precursors. The physical-chemical background, structure-retention relationships and optimization of separation of porphyrins and porphyrinogens by reversed-phase HPLC are discussed. There are practical tips based on application-oriented examples, including detailed instructions and protocols, from sample collection, handling and storage preparation, and HPLC separation and quantitation. Methods for measurement of haem biosynthetic enzyme activities are described, and characteristic porphyrin profiles in the blood, urine and faeces of patients with different types of porphyrias are given.
